RB2 is a fantasy football (FFB) position that refers to a team's second starting running back. The acronym is only used in FFB leagues that allow two or more RBs to start in the lineup.
Most FFLs feature starting lineups with 1 QB, 2 RBs, 3 WRs (or 2 WRs and a flex position), 1 TE, 1 K, and 1 team defense and special teams. RB1 and RB2 are often the most productive positions in leagues that implement standard scoring. However, PPR leagues give the WR positions added value because each catch equals a point (more or less than one point depending on league settings). The RB position can catch balls too though, which helps them retain value in PPR leagues.
